有直接的影響,但是可上調(diào)顆粒細(xì)胞中CYP2A4 和CYP19A1 的表達(dá),進(jìn)而促進(jìn)顆粒細(xì)胞中E2 和孕酮的
分泌,促進(jìn)卵巢的發(fā)育[19-20]。本試驗(yàn)發(fā)現(xiàn)高濃度褪黑素的添加在24h時(shí)可以顯著增強(qiáng)BMP6 的表達(dá),之
后會(huì)呈現(xiàn)抑制的現(xiàn)象可能是存在機(jī)體自身的負(fù)反饋調(diào)節(jié),72h后,高濃度(10-3 mol/L)和低濃度褪黑素
(10-11 mol/L)對(duì)BMP6 的表達(dá)沒有影響,其他濃度均顯著促進(jìn)其表達(dá)。
本研究驗(yàn)證了褪黑素對(duì)于卵巢顆粒細(xì)胞發(fā)育相關(guān)基因的影響,結(jié)果表明,不同濃度的褪黑素可以通過
促進(jìn)抗氧化相關(guān)基因(GPX4、SOD1)、發(fā)育相關(guān)基因(EGFR、DNMT1a)和TGFβ家族(BMP6)基因的表
達(dá),來減少氧化應(yīng)激對(duì)顆粒細(xì)胞功能的影響,刺激顆粒細(xì)胞的發(fā)育成熟,同時(shí)抑制FSHR 的表達(dá),保護(hù)顆
粒細(xì)胞抑制其凋亡。
